A differential scanning calorimetric study on the irreversible thermal unfolding of concanavalin A

چکیده
A differential scanning calorimetry study on the thermal denaturation of concanavalin A at pH 5.2 where it exists in the dimeric form was carried out. The calorimetric transitions were observed to be irreversible and the transition temperature of the protein increased with increasing scan rate, indicating that the thermal denaturation process is under kinetic control. The thermal unfolding, and its scan rate dependence could be explained according to the kinetic scheme N2 −→k 2U with k as first-order kinetic constant whose change with temperature is given by the Arrhenius equation. Using this model, rate constant as a function of temperature and activation energy of the process have been calculated. The average activation energy of the kinetic process using different approaches is 129±10 kJ mol−1. The differential scanning calorimetric results on transition temperatures and calorimetric enthalpies supported by intrinsic fluorescence indicate that the irreversibility in the calorimetric transitions of concanavalin A includes a combination of post-transition aggregation, chain separation and loss of cofactor.

Irreversible Denaturation of Maltodextrin Glucosidase Studied by Differential Scanning Calorimetry, Circular Dichroism, and Turbidity Measurements
Thermal denaturation of Escherichia coli maltodextrin glucosidase was studied by differential scanning calorimetry, circular dichroism (230 nm), and UV-absorption measurements (340 nm), which were respectively used to monitor heat absorption, conformational unfolding, and the production of solution turbidity. The denaturation was irreversible, and the thermal transition recorded at scan rates o...

High-sensitivity differential scanning calorimetry has been applied to the study of the thermal denaturation of thermolysin from Bacillus thermoproteolyticus rokko and its 255-316 and 205316 fragments. Stability investigations into thermolysin have been extended from a previous calorimetric study at pH 7.5 [2] to different experimental conditions, which included 0.3-3.7 mg/ml of protein concent...
متن کاملDifferential-scanning-calorimetric study of the irreversible thermal denaturation of 8 kDa cytotoxin from the sea anemone Radianthus macrodactylus.
A differential-scanning-calorimetric study of the thermal denaturation of a sea-anemone (Radianthus macrodactylus) 8 kDa cytolytic toxin was carried out. The calorimetric traces were found to be irreversible and scan-rate-dependent under the experimental conditions employed. Scan-rate-dependent thermograms were explained in terms of a two-state kinetic model N k -->D, where k is a first-order k...
متن کاملStability parameters for one-step mechanism of irreversible protein denaturation: a method based on nonlinear regression of calorimetric peaks with nonzero deltaCp.
Thermal transitions of many proteins have been found to be calorimetrically irreversible and scan-rate dependent. Calorimetric determinations of stability parameters of proteins which unfold irreversibly according to a first-order kinetic scheme have been reported. These methods require the approximation that the increase in heat capacity upon denaturation deltaCp is zero. A method to obtain th...

Bovine plasma proteins provide the needed amino acids for the growth and development of an organism. With the purpose of preserving the native structure, related with the protein functional properties, the oligosaccharide inulin was used as protective agent and was compared with glucose and sucrose, during freeze-drying. In the present study, the thermal stability of protein was investigated as...
